I think, to test world map path opener by SMBX, you must to remove your game save and start it from scratch, OR use PGE Engine's world map tester where you can type the exit code to trigger the right path opener that you have made on your map.
The setup you have made means, right path will be opened only when you will exit level by taking SMB3 Card roulette exit. If you got different exit way, this right path will NOT be opened.
